Serial dilution is the process by which a concentration of cells in a stock culture is made less concentrated with the addition of a diluent in various steps. <\/span><\/span><\/li>\n\t
The result is decreasing the number of cells in the original culture by an exponentially growing factor. <\/span><\/span><\/li>\n\t

Once diluted, you can then plate the culture to grow on solid agar media<\/span><\/li>\n<\/ul>\n\n
Two techniques are used:<\/span><\/p>\n\n

Spread plate: Agar is poured before and allowed to solidify; Then a volume of the diluted cell suspension is added to agar and spread with a spreader.<\/span><\/li>\n\t
Pour Plate: A volume of the diluted cell suspension is added to the petri plate and the warm agar(45-50oC) is then poured into the plate.
